Synthesis of Silver Nanoparticles, Study of Structure, Properties and Applications in the Treatment of Wounds

Silver nanoparticles are used in many areas of medicine as part of antibacterial and wound-healing ointments and preparations. Research objectives: study the synthesis of silver nanoparticles, structure, properties and applications in the treatment of burn wounds. The synthesis of silver nanoparticles was performed by chemical precipitation. As synthesized silver nanoparticles have a cubic crystalline shape; the average size of nanoparticles is 18 nm. No skin-irritating effect of medicinal cosmetic gel containing silver nanoparticles was detected.

Development of CuO Nanoparticles from the Mucus of Garden Snail Cornu aspersum as New Antimicrobial Agents

Several biologically active compounds involved in the green synthesis of silver and gold nanoparticles have been isolated from snail mucus and characterized. This paper presents a successful method for the application of snail mucus from Cornu aspersum as a bioreducing agent of copper sulfate and as a biostabilizer of the copper oxide nanoparticles (CuONPs-Muc) obtained. The synthesis at room temperature and neutral pH yielded nanoparticles with a spherical shape and an average diameter of 150 nm. The structure and properties of CuONPs-Muc were characterized using various methods and techniques, such as ultraviolet–visible spectroscopy (UV–vis), high-performance liquid chromatography (HPLC), one-dimensional polyacrylamide gel electrophoresis (1D-PAGE), up-conversion infrared spectroscopy Fourier transform (FTIR), scanning electron microscopy combined with energy dispersive spectroscopy (SEM/EDS), Raman spectroscopy and imaging, thermogravimetric analysis (TG-DSC), etc. Mucus proteins with molecular weights of 30.691 kDa and 26.549 kDa were identified, which are involved in the biogenic production of CuONPs-Muc. The macromolecular shell of proteins formed around the copper ions contributes to a higher efficiency of the synthesized CuONPs-Muc in inhibiting the bacterial growth of several Gram-positive (Bacillus subtilis NBIMCC2353, Bacillus spizizenii ATCC 6633, Staphylococcus aureus ATCC 6538, Listeria innocua NBIMCC8755) and Gram-negative (Escherichia coli ATCC8739, Salmonella enteitidis NBIMCC8691, Salmonella typhimurium ATCC 14028, Stenotrophomonas maltophilia ATCC 17666) bacteria compared to baseline mucus. The bioorganic synthesis of snail mucus presented here provides CuONPs-Muc with a highly pronounced antimicrobial effect. These results will expand knowledge in the field of natural nanomaterials and their role in emerging dosage forms.

Microwave-assisted green synthesis of silver nanoparticles using turnip root (Brassica rapa subsp. Rapa) extract and their antibacterial efficacy

Biosynthesised silver nanoparticles (AgNPs), especially those synthesised by plant extracts, are extensively utilised in the fields of pharmacy and medicine due to their ability to exhibit a wide range of biological functions. The current study is focused on the green synthesis of silver nanoparticles from various concentrated silver nitrate solutions using turnip root extract as a reducing and stabilising agent and to assess the in vitro antibacterial efficacy of these AgNPs. For the study, we employed microwave-assisted reduction to examine the impact of microwave energy on the resulting AgNPs in comparison to the conventional heating method. X-ray diffraction analysis revealed the crystalline nature of the as-prepared AgNPs, while field emission-scanning electron microscopy (FE-SEM) showed the majority of spherical-shaped AgNPs had an average grain size of 47.5 nm. Energy-dispersive x-ray spectroscopy (EDX) indicates the high weight percentage of the produced AgNPs than for extract residue, especially for the AgNPs that are prepared with microwave assistance. Fourier transform infrared spectroscopy (FTIR) was used to reveal the functional groups that are related to many phytochemical compounds, such as alkaloids, flavonoids, and terpenoids, which act as reducing and stabilizing agents. High zeta potential measures (−22.77 and −38.83 mv) approved the high stability of the synthesised AgNPs as higher zeta potentials typically correlate with greater stability. The antibacterial activity behaviour of the produced NPs against gram-positive (Staphylococcus aureus) and gram-negative (Escherichia coli) bacteria was significantly acceptable for both heating techniques, as observed from the measured inhibition zone.

‘GREEN METAL NANO TECHNOLOGY IN DENTISTRY’-A NARRATIVE REVIEW

Over time, there has been a lot of interest in the synthesis of green AgNPs utilising plant extracts due to the remarkable antibacterial capabilities of these nanoparticles. These particles' nanosized makes it easier for them to pass through cell membranes, interacts with internal organelles, and eventually damages cells when they proliferate. With the exceptional benefits, including practicality, simplicity of usage, and the utilisation of specialised processes like bacteria, plants are currently the preferred method for synthesising metal nanoparticles. Based on the idea of converting silver ions (Ag+) to silver nanoparticles (Ag0), green synthesis techniques for the synthesis of silver nanoparticles are provided. The literature reveals that the modified metal nano particles modifying with green methods are already practicing by dental clinicians, whereas still less percentage of practitioners are using this green nano technology. Hence this narrative review includes different metal nano particles using in dentistry and different green methods using to modify the environmentally friendly dental practices.

Biofabrication of Silver Nanoparticles by Azadirachta indica Rhizosphere Bacteria with Enhanced Antibacterial Properties

Microorganisms (MOs) are prominent in ecological functioning and balance. The rhizosphere is considered one of the most diverse ecosystems on Earth and serves as a breeding spot for many MOs. Rhizosphere microbial diversity changes according to plant species, genotype, and the nature of the soil. The current study reports the possible use of bacteria isolated from the rhizosphere of Azadirachta indica for synthesizing silver nanoparticles (AgNPs). The physicochemical characterization and antibacterial activity of these green synthesized AgNPs are also reported. The gene (16S rRNA) sequence of bacteria isolated from the rhizosphere showed a maximum similarity of 99.25% with Bacillus subtilis. After incubation, the colorless reaction mixture transformed to brown, which indicates the formation of AgNPs, and UV-vis spectral analysis also confirmed the biosynthesis of AgNPs. Compared to lower temperatures, the efficiency of AgNP synthesis was high at the higher temperature. The scanning electron microscope image demonstrated spherical-shaped AgNPs with sizes ranging from 18 to 21 nm. Energy-dispersive X-ray analysis established the elemental analysis of synthesized AgNPs. The synthesized AgNPs showed strong bactericidal properties against pathogenic bacteria Klebsiella pneumonia, Pseudomonas aeruginosa, Escherichia coli, and methicillin-resistant Staphylococcus aureus.

Entomopathogenic fungi-based silver nanoparticles: a potential substitute of synthetic insecticides to counter behavioral and physiological immunity in Aedes aegypti mosquito (Diptera: Culicidae).

Excessive use of synthetic insecticides has resulted in environmental contamination and adverse effects on humans and other non-target organisms. Entomopathogenic fungi offer eco-friendly alternatives; however, their application for pest control requires significant advancement owing to limitations like slow killing time and effectiveness only when applied in higher amounts, whereas exposure to UV radiation, high temperature, and humidity can also reduce their viability and shelf-life. The nanoparticles synthesized using fungal extracellular extracts provide a new approach to use fungal pathogens. Our study focused on the synthesis of Metarhizium anisopliae-based silver nanoparticles (AgNPs) and evaluation of their efficiency on various physiological and behavioral parameters of the mosquito Aedes aegypti. The synthesis, size (27.6 d.nm, PDI = 0.209), zeta potential (- 24.3 mV), and shape of the AgNPs were determined through dynamic light scattering, scanning and transmission electron microscopic, and UV-visual spectroscopic analyses (432 nm). Our results showed significantly reduced survival (100% decrease in case of 3.2 and 1.8 μL/cm2 volumes, and 60% decrease in case of 0.8 μL/cm2 volume), phenoloxidase activity (t = 39.91; p = 0.0001), and gut microbiota, with increased oxidative stress and cell apoptosis in AgNPs-challenged mosquitoes. Furthermore, the AgNPs-exposed mosquitoes presented a concentration-specific decrease in flight locomotor activity (F = 17.312; p < 0.0001), whereas no significant changes in antifungal activity, self-grooming frequencies, or time spent were found. These findings enhance our understanding of mosquito responses to AgNPs exposure, and offer a more efficient mosquito control strategy using entomopathogenic fungi.

Altaf et al. recently published in Materials, 2022; 15(11), 3826, about the synthesis of silver nanoparticles (AgNPs) using the non-thermal plasma reduction of AgNO3 salt and performed statistical optimization for the reaction conditions, i.e., (A) the concentration of a stabilizing agent, mM (B) concentration of AgNO3 salt, mM and (C) the reaction time, mins. We would like to point out that their writing on the statistical analysis (Box–Behnken response surface methodology for predicting the size of the nanoparticles) is not complete and, therefore, cannot be independently checked by the readers. The problems found in their report are as follows: the hard-to-find actual value of the uncoded units; a dubious claim about the middle levels of variable B (salt concentration); inconsistency in using coded vs. uncoded units in the table vs. the regression equation; and three center points with identical conditions give a dissimilar prediction of results. These serious issues need to be clarified and revised, as well as several writing errors, in order to uphold the standard of scientific publications.

Synthesis, Characterization, and Antibacterial Activity of Silver Nanoparticles Supported on Bovine Bone Powder Using Heterotheca inuloides: An In Vitro Study

This paper reports on the biosynthesis, characterization, as well as the bactericide and cytotoxic properties of silver nanoparticles supported on bovine bone powder (Ag-NPs/BBP). The silver nanoparticles were obtained through the bioreduction of AgNO3, using an infusion of Heterotheca inuloides leaves and flowers as a reducing agent and bovine bone powder as a support. The ratio of Ag-NPs/bovine bone powder was set as 1:10. The characterization was performed with SEM–EDS, XRD, UV–Vis, and TEM, which showed the formation of nanoparticles with an average size of 22.6 ± 10.8 nm and a quasi-spherical Ag-NPs morphology supported on the BBP surface. The nanocomposite exhibited a band gap of 2.19 eV. The minimal inhibitory concentration and the minimal bactericidal concentration against S. aureus, E. coli, and S. epidermidis were determined for each strain. In addition, the cytotoxic evaluation of the Ag-NPs/BBP on J774.2 mouse macrophage cells was performed. The Ag-NPs/BBP exhibited a bactericide effect on the strains studied, and the cytotoxicity had a dose-dependent behavior on the cells studied. Therefore, it was found that the ecofriendly synthesized Ag-NPs supported on bovine bone powder resulted in an effective bactericidal system against the strains studied, without significant cytotoxicity.

Development of phytosterol-loaded silver nanoparticles for ameliorating haemorrhoidal complications via the AMPK pathway—a mechanistic approach

The aim of the current study was to synthesize silver nanoparticles (PLSNPs) using green technology by means of phytosterol-enriched fractions from Blumea lacera extracts (EAF) and evaluate their toxicological and anti-haemorrhoidal potential. The average size of the synthesized particles was found to be 85.64 nm by scanning electron microscopy and transmission electron microscopy. Energy dispersive spectroscopy showed the elemental composition of PLSNPs to be 12.59% carbon and 87.41% silver, indicating the capping of phytochemicals on the PLSNPs. The PLSNPs were also standardized for total phytosterol content using chemical methods and high-perfromance liquid chromatography. The PLSNPs were found to be safe up to 1000 mg kg−1 as no toxicity was observed in the acute and sub-acute toxicity studies performed as per OECD guidelines. After the induction of haemorrhoids, experimental animals were treated with different doses of EAF, PLSNPs and a standard drug (Pilex) for 7 d, and on the eighth day the ameliorative potential was assessed by evaluating the haemorrhoidal (inflammatory severity index, recto-anal coefficient) and biochemical (tumour necrosis factor-alpha and interleukin-6) parameters and histology of the recto-anal tissue. The results showed that treatment with PLSNPs and Pilex significantly (p < 0.05) reduced haemorrhoidal and biochemical parameters. This was further supported by restoration of altered antioxidant status. Further, a marked reduction in the inflammatory zones along with minimal dilated blood vessels was observed in the histopathological study. The results of molecular docking studies also confirmed the amelioration of haemorrhoids via AMP-activated protein kinase (AMPK)-mediated reduction of inflammation and endothelin B receptor modification by PLSNPs. In conclusion, PLSNPs could be a good alternative for the management of haemorrhoids.

Deformed graphene FET biosensor on textured glass coupled with dielectrophoretic trapping for ultrasensitive detection of GFAP.

Numerous efforts have been undertaken to mitigate the Debye screening effect of FET biosensors for achieving higher sensitivity. There are few reports that show sub-femtomolar detection of biomolecules by FET mechanisms but they either suffer from significant background noise or lack robust control. In this aspect, deformed/crumpled graphene has been recently deployed by other researchers for various biomolecule detection like DNA, COVID-19 spike proteins and immunity markers like IL-6 at sub-femtomolar levels. However, the chemical vapor deposition (CVD) approach for graphene fabrication suffers from various surface contamination while the transfer process induces structural defects. In this paper, an alternative fabrication methodology has been proposed where glass substrate has been initially texturized by wet chemical etching through the sacrificial layer of synthesized silver nanoparticles, obtained by annealing of thin silver films leading to solid state dewetting. Graphene has been subsequently deposited by thermal reduction technique from graphene oxide solution. The resulting deformed graphene structure exhibits higher sensor response towards glial fibrillary acidic protein (GFAP) detection with respect to flat graphene owing to the combined effect of reduced Debye screening and higher surface area for receptor immobilization. Additionally, another interesting aspect of the reported work lies in the biomolecule capture by dielectrophoretic (DEP) transport on the crests of the convex surfaces of graphene in a coplanar gated topology structure which has resulted in 10 aM and 28 aM detection limits of GFAP in buffer and undiluted plasma respectively, within 15 minutes of application of analyte. The detection limit in buffer is almost four decades lower than that documented for GFAP using biosensors which is is expected to pave way for advancing graphene FET based sensors towards ultrasensitive point-of-care diagnosis of GFAP, a biomarker for traumatic brain injury.

Green Approach for Synthesis of Silver Nanoparticles with Antimicrobial and Antioxidant Properties from Grapevine Waste Extracts

The present work aims to study the possibilities of developing silver nanoparticles using natural extracts of grape pomace wastes originating from the native variety of Fetească Neagră 6 Șt. This study focused on investigating the influence of grape pomace extract obtained by two different extraction methods (classical temperature extraction and microwave-assisted extraction) in the phytosynthesis process of metal nanoparticles. The total phenolic content of the extracts was assessed using the spectrophotometric method with the Folin–Ciocâlteu reagent, while the identification and quantification of specific components were conducted through high-performance liquid chromatography with a diode array detector (HPLC-DAD). The obtained nanoparticles were characterized by UV–Vis absorption spectroscopy, X-ray diffraction (XRD), and transmission electron microscopy (TEM), along with assessing their antioxidant and antimicrobial properties against Gram-positive bacteria. The data collected from the experiments indicated that the nanoparticles were formed in a relatively short period of time (96 h) and, for the experimental variant involving the use of a 1:1 ratio (v/v, grape pomace extract: silver nitrate) for the nanoparticle phytosynthesis, the smallest crystallite sizes (from X-ray diffraction—4.58 nm and 5.14 nm) as well as spherical or semispherical nanoparticles with the lowest average diameters were obtained (19.99–23 nm, from TEM analysis). The phytosynthesis process was shown to enhance the antioxidant properties (determined using the DPPH assay) and the antimicrobial potential (tested against Gram-positive strains) of the nanoparticles, as evidenced by comparing their properties with those of the parent extracts; at the same time, the nanoparticles exhibited a selectivity in action, being active against the Staphylococcus aureus strain while presenting no antimicrobial potential against the Enterococcus faecalis strain.

A Silver Nanoparticle‐Embedded Tamarind Kernel Gum/Poly (Sodium Acrylate) Nanocomposite for Sustainable Release of Doxycycline

AbstractThe current work describes an eco‐friendly novel synthesis of silver (Ag) nanoparticles (NPs)/Tamarind kernel gum (TKG)/Poly (sodium acrylate) (PSA) nanocomposite without the use of any toxic materials. It is a one‐step route synthesis of silver nanocomposite hydrogel in which TKG and PSA are cross‐linked using methylene bisacrylamide (MBA) cross‐linker. The developed hydrogels were characterized using XRD, FTIR, UV‐visible, FE‐SEM, and TEM techniques. Doxycycline (dox) was chosen as a model drug for release studies using TKG/PSA and Ag/TKG/PSA nanocomposite. The observation is that Ag NP/TKG/PSA nanocomposite showed a pH‐dependent swelling which is higher at pH 7.4 than at pH 1.2. Similarly, the cumulative drug release percentage (CDR %) using silver nanocomposite is higher at pH 7.4 than at pH 1.2. Also, the presence of Ag NPs decreased the swelling ratio (SR) of TKG/PSA hydrogel, which controls the rate of release of the dox drug. In addition, the dox release kinetics were studied using the Korsmeyer‐Peppas and Higuchi model. It has been found that drug release data was fitted in the Korsmeyer‐Peppas model with a higher value of regression coefficient (R2). Thus, Ag/TKG/PSA nanocomposite showed satisfactory results for sustainable release of doxycycline drug.

Synthesis of bio-stabilized silver nanoparticles using Roccella montagnei, their anticandidal capacities & potential to inhibit the virulence factors in fluconazole-resistant Candida albicans.

Candida species is the causative agent in approximately 80% of invasive mycoses and drug-resistant Candida albicans is among the four strains of 'critical priority group' framed by WHO. Lichens are endowed with some rare phytochemicals and a plethora of therapeutics viz. antifungal capacities of Roccella montagnei. Biosynthesis of silver nanoparticles (AgNPs) using lichen could offer an eco-friendly, and cost-effective alternative against emerging 'microbial resistance.' Therefore, the objective was to biosynthesize silver nanoparticles (Rm-AgNPs) using a Hydro-alcoholic (1:1) extract of R. montagnei to develop a potent anticandidal agent against Fluconazole-resistant C. albicans NBC099. UV-Spectroscopy identified AgNPs specific-peak of Rm-AgNPs at 420-440nm and FTIR revealed the presence of amines, alcohol, aromatic compounds, and acids. SEM and TEM analysis indicated that Rm-AgNPs are spherical shaped with a size range of 10-50nm. Zetasizer analysis indicated that particles are highly stable and have a mean hydrodynamic diameter of 116nm with a zeta potential charge of - 41mV. XRD analysis suggested face centered cubic crystal lattice structure. Results indicated that Rm-AgNPs strongly inhibited the growth of NBC099 at a minimum inhibitory concentration (IC50) of ≤ 15µg. C. albicans culture treated with Rm-AgNPs at concentrations below IC50, down-regulates the production of different virulence factors in NBC099, viz. hyphal formation (> 85%), biofilms production (> 80%), phospholipase, esterase, proteinase activity. The apoptosis assay demonstrated the Rm-AgNPs induced apoptosis in NBC099 cells via oxidative stress. Interestingly, Rm-AgNPs showed negligible cytotoxicity (< 6%) in murine RAW 246.7 macrophage cells at a concentration above 15µg/mL. Therefore, Rm-AgNPs have been offered as an anti-candida alternative that can be utilized to improve the efficacy of already available medications.

Catalytic Degradation Efficacy of Silver Nanoparticles Fabricated Using Actinidia deliciosa Peel Extract

The preparation of metallic nanoparticles using green synthetic approaches and its application toward the efficient degradation of environmentally hazardous dyes constitutes an attractive alternative to currently employed methods. In the current report, the green synthesis of silver nanoparticles (AgNPs) was successfully achieved using Actinidia deliciosa (kiwifruit) peel aqueous extract as a bioreducing agent under optimized synthesis conditions. The experimental parameters were optimized in terms of reactant ratio, reaction temperature, and reaction time. The biogenic nanoparticles exhibited SPR absorption band at λmax 480 nm. Transmission electron microscopy (TEM) and scanning electron microscopy (SEM) images revealed quasispherical monodisperse nanoparticles which were 36 nm in diameter. The hydrodynamic diameter of the nanoparticles was 106 nm as determined by dynamic light scattering, and the highly negative ζ-potential (−34 mV) supported its superior colloidal stability. Energy dispersive X-ray confirmed that silver is a major constituent of the nanoparticles. X-ray diffraction (XRD) diffractograms confirmed the crystallinity of the nanoparticles and its face-centered cubic (fcc) lattice structure. The functional groups in the plant’s phytochemicals facilitating the reduction of Ag+ ions and stabilization of the formed AgNPs were identified by fourier transform infrared (FTIR) spectroscopy. In specific, the bands in the FTIR spectra at 3,412, 1,618, 1,419, and 1,237 cm−1 suggested the presence of phenolic compounds. Phytochemical analysis by colorimetric assays revealed that the kiwifruit peel extract was rich in phenolic compounds. When evaluated in the catalytic degradation of organic dyes, the biosynthesized AgNPs induced instant and complete discoloration of the methylene blue dye when 1.6 mg of nanoparticles was used. At a lower dose of AgNPs (0.4 mg), 80% degradation of the dye occurred after 3 hr of treatment. The degradation reaction followed second-order kinetics with a rate constant of 0.01083 mM−1s−1. The current study highlights the immense potential of the prepared nanoparticles as efficient catalysts for the degradation of hazardous organic dyes such as methylene blue and presents an intriguing argument for investigating the catalytic efficiency of the biogenic AgNPs for the degradation of other structurally different dye pollutants.

Green synthesis approach: Utilizing chrysanthemum extract as reducing and stabilizing agent for the fabrication of silver nanoparticles and their antimicrobial properties study

This research explores the eco-friendly synthesis of silver nanoparticles (AgNPs) using Chrysanthemum extract, emphasizing its application in antimicrobial treatments. The study primarily focused on assessing the effect of extract concentration on nanoparticle characteristics. The synthesized AgNPs, at higher Chrysanthemum extract concentrations (up to 15%), exhibited a notable decrease in size, from 48.5 nm at 2% to 22.1 nm, and an increase in yield, achieving 2.24 mg/mL. The AgNPs showed a significant antibacterial effect, with the lowest IC50 value of 2.08 µg/mL against Vibrio cholerae, demonstrating a broad-spectrum efficacy. Advanced characterization techniques like SEM and FTIR spectroscopy confirmed the morphological integrity and phytochemical capping of the nanoparticles. The study successfully highlights the potential of Chrysanthemum extract not only as a sustainable source for nanoparticle synthesis but also in enhancing the biocompatibility and efficacy of AgNPs, paving the way for their application in medical and environmental fields.

Phyto-fabrication of Moringa oleifera peel-sourced silver nanoparticles: A promising approach for combating hepatic cancer by targeting proinflammatory cytokines and mitigating cytokine storms.

Hepatocellular carcinoma (HCC) arises from precancerous nodules, leading to liver damage and inflammation, which triggers the release of proinflammatory cytokines. Dysregulation of these cytokines can escalate into a cytokine storm, causing severe organ damage. Interestingly, Moringa oleifera (M. oleifera) fruit peel, previously discarded as waste, contains an abundance of essential biomolecules and high nutritional value. This study focuses on the eco-friendly synthesis of silver nanoparticles infused with M. oleifera peel extract biomolecules and their impact on regulating proinflammatory cytokines, as well as their potential anticancer effects against Wistar rats. The freshly synthesized nanoformulation underwent comprehensive characterization, followed by antihepatic cancer evaluation using a diethyl nitrosamine-induced model (at a dose of 200 mg kg-1 BW).
